Human and mouse neurons don’t keep herpes dormant the same way

باختصارA viral protein called ICP34.5 turned out to help HSV‑1 stay dormant in human neurons grown from stem cells, but it played a different role in mouse neurons. Results from mice don’t always carry over to people, and better human models are needed.

To cure herpes, scientists need to understand how the virus stays dormant (latent) and what makes it wake up (reactivate). Much of that knowledge comes from mice. This study from David Leib’s lab at Dartmouth, published in Virology, compared mouse neurons with human sensory neurons made from stem cells.

What ICP34.5 does

ICP34.5 is an HSV‑1 protein that helps the virus get around the cell’s built-in defenses, including the interferon response and the shutdown of protein production that infected cells use to stop viruses.

What they found

  • In human neurons, ICP34.5 wasn’t needed for the virus to become dormant, but it was important for keeping it dormant: virus lacking ICP34.5 reactivated on its own more often.
  • In human neurons, its main effect came through controlling the innate immune (interferon) response, rather than autophagy, the cell’s recycling system.
  • In mouse neurons, ICP34.5 mattered mostly when reactivation was triggered, and the host’s interferon response was the key to keeping the virus dormant, with or without ICP34.5.

Why it matters

The same viral protein behaved differently in human and mouse neurons. That is a reminder that results from mice don’t always predict what happens in people, and that human-cell models are becoming an important step before testing treatments that target latency.

What to keep in mind

This is basic laboratory research in cultured neurons. It doesn’t point to a treatment yet; it improves the map that future treatments will rely on.
